Crisaborole

Indications

Crisaborole is prescribed for the treatment of mild to moderate atopic dermatitis in children aged three months and up.

Pharmacology

Crisaborole, the active component, is a phosphodiesterase-4 (PDE-4) inhibitor that works primarily on the inflammatory phosphodiesterase 4B (PDE4B). Crisaborole is a phenoxybenzoxaborole chemically. It contains a boron atom, which aids in skin penetration and is required for binding action. PDE4B inhibition appears to reduce the production of cytokines such as tumor necrosis factor alpha (TNF), interleukin-12 (IL-12), IL-23, and other proteins involved in the immune response and inflammation.

Interaction

Crisaborole and metabolite 1 are not likely to inhibit cytochrome P450 (CYP) 1A2, 2B6, 2C8, 2C9, 2C19, 2D6, and 3A4 under clinical settings, according to in vitro experiments utilizing human liver microsomes. In vitro tests on metabolite 2 in human liver microsomes revealed that it did not impede the activities of CYP2C19, 2D6, or 3A4.

Side Effects

Crisaborole has the potential to trigger allergic responses at or around the application site. Hives, itching, swelling, and redness are all symptoms that might be dangerous. Crisaborole's most common side effect is pain at the application site, such as burning or stinging.

Pregnancy & Lactation

Pregnancy: No evidence on Crisaborole in pregnant women is available to determine the drug's risk of serious birth problems or miscarriage. In animal reproduction experiments, oral administration of crisaborole to pregnant rats and rabbits during organogenesis had no adverse developmental effects at dosages up to 3 and 2 times, respectively, the maximum recommended human dose (MRHD).

Lactation: No information on the presence of crisaborole in human milk is known. Crisaborole is absorbed throughout the body. A firm determination of the risk of crisaborole to a breastfed newborn is impossible due to a lack of clinical data during breastfeeding.

Precautions & Warnings

Patients treated with Crisaborole have experienced hypersensitivity responses, including contact urticaria. If signs and symptoms of hypersensitivity appear, stop taking Crisaborole right once and seek medical help.
